Entity

DNA polymerase III

Enzyme that synthesizes most of the new DNA during bacterial replication.

It is the main enzyme that synthesizes new DNA during bacterial replication, adding nucleotides complementary to the template strand in the 5′ to 3′ direction.

It is part of a multiprotein complex, the replisome, and works together with a sliding-clamp processivity subunit that keeps it attached to the DNA. It always requires a primer to start synthesis, since it cannot begin a strand de novo.

The video explains that it needs the primer placed by primase to start adding nucleotides to the template strand, synthesizing the leading strand continuously and the lagging strand in Okazaki fragments, since it needs a new primer each time.
